In flowering plants, crosses between parents with differing chromosome numbers often fail to develop viable seeds because of unbalanced expression of imprinted genes in the endosperm. This post-zygotic hybridization barrier is known as the "triploid block" response, and has been regarded as a major path towards reproductive isolation and polyploid speciation. However, the natural forces defining its strength and evolutionary impact in wild populations have not been fully investigated. Previous results have shown that plants use DNA methylation and transposon-derived small RNA to sense parental genome dosage in the endosperm of hybrid seeds. The paternal epigenome is a key component of this process, as loss of DNA methylation and small-interfering RNAs in diploid pollen allows bypassing the triploid block almost completely. In this seminar I will present our current efforts to identify the origins and dynamics of reproductive small RNAs responsible for hybrid seed collapse, as well as their targets.
